In detail, 5 [micro]g of total RNA was pooled from four individual animals within the same breed and then double-stranded cDNA containing the T7 RNA polymerase promoter sequence (5'-AAACG ACGGC CAGTG AATTG T AATA CGACT CACTA TAGGC GC-3') was synthesized using a TaKaRa cDNA Synthesis System Kit according to manufacturer's recommended protocol (TaKaRa, Dalian, China).
