For [beta]-galactosidase activity determination, 20 [micro]L of a 4 mg/mL solution of chlorophenol red-[beta]-D-galactopyranoside (CPRG; Sigma) and 150 [micro]L CPRG buffer (60 mM [Na. After 24-48 hr, the absorbance of the red color due to the hydrolysis product of CPRG was read using a microplate reader (Model 550; Bio-Rad Laboratories, Hercules, CA, USA) at 540 nm. CPRG is converted to chlorophenol red and reflects [Beta]-galactosidase activity, which is indicative of the estrogenic activity. |
