The enzymatic reaction was stopped by addition of 50 [micro]L of chromophore reagent (40 mmol/L DBPDA in 6 mol/L HCI), followed by addition of 50 [micro]L of oxidizing agent (40 mmol/L potassium ferricyanide in 20 mmol/L potassium phosphate buffer, pH 8. 2]S combines with DBPDA to form 3,7-bis(dibutyl amino)phenothiazine-5-ium chloride, which is highly fluorescent. |
