In the absence of functional p53 protein, cells lose genomic integrity and are prone to cancer development, p53 functions as a transcription factor up-regulating many target genes, such as p21 (cydin-dependent kinase inhibitor 1A, Cipl), MDM2 (transformed 3T3 cell double minute 2), and GADD45 (growth arrest and DNA-damage-inducible, alpha) (Levine 1997). DNA damage-induced phosphorylation of p53 alleviates inhibition by MDM2. |
