Histologic and in situ hybridization analyses of livers from cynomolgus macaques infected with Lassa virus (LASV) (1 x [10.sup.4] 50% tissue culture infective dose [[TCID.sub.50]]) and treated 1 time daily with favipiravir (treated) or vehicle only (control).
Samples that tested positive for IgG antibodies to SARS-CoV were further assayed for neutralizing antibodies by using the 50% tissue culture infective dose (TCI[D.sub.50]) method, similar to that previously described (6), under biosafety level 3 conditions, in serial twofold dilution, ranging from 1:10 to 1:320.
In this case, the virus stocks ([10.sup.-4.2] 50% tissue culture infective dose) were diluted 1:400, and virus neutralization titers were checked 3 days later.
For example, no decrease was observed in a 50% tissue culture infective dose (TCI[D.sub.50]) of poliovirus samples after they were mounted on the grid and stained with 2.5 mM (1.6%) phosphotungstic acid, pH 7.0.
The recognized sensitivity (0.01 50% tissue culture infective dose) and specificity of our enterovirus-RNA detection assay allowed us to detect specific enterovirus RNA sequences in 46.3% (25 of 54) of CSF specimens.
AWN virus control was prepared by extracting RNA from a 100-[micro]L volume containing 10 50% tissue culture infective dose WN stock virus.
