We assayed adiponectin in whole milk and leptin, AFABP, and EFABP in skim milk.
We performed all determinations of adiponectin, leptin, and AFABP with commercially available ELISA kits (Biovendor-Laboratory Medicine).
To validate the assay, 2 skim breast milk samples with the baseline AFABP concentrations of 8.4 and 6.5 [micro]g/L were enriched with various amounts of AFABP calibrator to increase the AFABP concentrations by +2.0 and +5.0 [micro]g/L.
No crossreactivity was found with recombinant human AFABP (Biovendor), IFABP (R&D Systems), LFABP (Abnova), or other related proteins: leptin, leptin receptor, adiponectin, resistin, and RELM-(3 at 100 [micro]g/L and IL-6 at 2 [micro]g/L (all provided by Biovendor).
We next measured the serum concentrations of AFABP in 100 lean and 129 overweight or obese individuals.
Notably, the correlations of serum AFABP with fat percentage and triglyceride, insulin, and 2-h postprandial glucose concentrations were significant even after adjustment for BMI.
Age- and sex-adjusted serum concentrations of AFABP were inversely associated with circulating concentrations of adiponectin, an important adipokine with antidiabetic and antiatherogenic activities (32).
