In this video, the Exo Bacto Scrub has been pitted against an ordinary scrubber.
Solidified media with bacto Agar achieved a higher germination percentage with lesser genetic variation.
After 48 h, shoot tips were transferred to a recovery medium (MS supplemented with 2 mg/L BAP and 7 g/L Bacto agar).
The isolates were inoculated into the conical flask containing Bacto Bushnell Hass broth in artificial sea water along with 2% crude oil and dextrose as additional carbon source [2].
Two types of agar, marine agar (Difco Marine Broth 2,216 and Bacto Agar) and nutrient agar (Peptone, 5 g; Yeast extract, 3 g; NaCl, 5 g; Agar, 15 g; Milli-Q water, 1,000 mL) were used for the recovery of aerobic bacteria.
The sediment of specimens was inoculated on Sabouraud dextrose agar, Sabouraud dextrose agar (Merck, Germany) with cycloheximide (Sigma-Aldrich, USA), paraffin agar (0.05 g FeS[O.sub.4], 0.05 g MgS[O.sub.4]x7[H.sub.2]O, 1 g [K.sub.2]HP[O.sub.4], 5 g N[H.sub.4]Cl, 0.05 g ZnS[O.sub.4], 1 g N[H.sub.4]N[O.sub.3], 0.05 g MnS[O.sub.4], 3 g K[H.sub.2]P[O.sub.4], 17 g Bacto Agar and 1 liter distilled water) and McClung's carbon-free broth tube (0.5 g MgS[O.sub.4]x7H, 2 mg ZnS[O.sub.4], 10 mg Fe[Cl.sub.3], 8 mg Mn[Cl.sub.2] x 4[H.sub.2]O, 0.8 g [K.sub.2]HP[O.sub.4], 2 g NaN[O.sub.3], 1 liter distilled water, pH 7.2) with placement of a paraffin coated glass rod.
They were cultured in 10 mL of tryptic soy broth (TSB; Bacto, Becton Dickinson, MD, USA), and incubated at 35[degrees]C for 2 h.
harveyi, which is an important pathogen of aquacultured Pacific white shrimp, was cultured at 30 [degrees]C in stirred suspension culture in sea-water complete culture medium (seawater, 750 ml; distilled water, 250 ml; Bacto tryptone, 5 g; yeast extract, 1 g; glycerol, 3 ml) and Difco Marine Broth 2216 (BD Biosci-ence) and was used when in log-phase growth.
