The nuclei were counterstained with hematoxylin and the sections were coverslipped with
DePex. As negative control the procedure was performed omitting the first antibody.
After serial dehydration and mounting with
Depex, the slides were examined with the aid of a Leica DMRE microscope equipped with a Leica 63 x/1.32-0.6 oil objective; images were acquired with a Leica DFC320 camera.

Finally, the cells were mounted with
DePeX (SERVA Electrophoresis, Heidelberg, Germany) and the production of GHS-R was assessed with an Axioskop 2 microscope (20x, Carl Zeiss, Germany).
The sections were counterstained with haematoxylin before being dehydrated in graded alcohol and xylene and mounted with
Depex (Leica Biosystems, USA).
The otolith sections were polished with wet and dry carborundum paper (grade 1200) and mounted on microscope slides with
DePeX mounting adhesive (VWR International Ltd., Poole, England) and a cover slip.
All slides were dehydrated in ethanol and mounted in a synthetic resin (
Depex, Serva, Heidelberg, Germany).
After dehydration with ethanol (960 mL/L ethanol followed by absolute ethanol), the sections were embedded with
DePeX mounting medium "Gurr" (BDH Laboratory Supplies).
The cover slips were dehydrated in 100% ethanol and xylol before slides were cover slipped with
DePex.
