In brief, translation initiation includes (1) binding of the GTP-bound form of eukaryotic initiation factor 2 (eIF2) and initiator methionyl-tRNA (met-tRN[A.sub.i]) complex to the small (40S) ribosomal subunit to form the 43S preinitiation complex (PIC), (2) binding of the 43S PIC to the 7-methylguanosine ([m.sup.7]G) cap of the 5' end of messenger RNA (mRNA), and (3) 43S PIC scanning of the mRNA for an AUG initiation codon [24, 25].
