Inicialmente, a alga, desidratada em temperatura ambiente (25[degrees]C) e macerada com [N.sub.2] liquido (5 g), foi hidratada em 250 mL de tampao acetato de sodio 0,1 M (AcNa) (pH 5,0) (Vetec Quimica), contendo EDTA 5 mM (QEEL
) e cisteina 5 mM (Sigma Chemical).
Firstly the ground material (5 g) was hydrated in 250 mL of 100 mM sodium acetate buffer (NaAc) (Vetec Quimica) (pH 5.0) containing EDTA (QEEL
) and cysteine (5 mM) (Sigma Chemical), and digested in a crude papain solution (30 mg [mL.sup.-1]) (Vetec Quimica) for 6 hours at 60[degrees]C in bain-marie (Marconi, model MA 159).
To that end, algae (5 g) dehydrated at room temperature and macerated with liquid [N.sub.2] were hydrated in 250 mL of 100 mM sodium acetate buffer (pH 5.0) (Vetec Quimica) containing 5 mM EDTA (QEEL
) and 5 mM cysteine (Sigma Chemical), and subjected to digestion with 17 mL of crude papain solution (30 mg [mL.sup.-1]) during 6 hours at 60[degrees]C in water bath (MARCONI, model MA 159).
