'Through the TCBS, Tolkien felt his first comradery in a group of all males, among friends dedicated to something higher than themselves,' writes Bradley J.
Then use the 4.5 mL sterilized seawater diluted to 10-1, 10-2, 10-3 and 100, 10-1, 10-2 concentrations for 2216E and TCBS plates, respectively.
To compare the aerodynamic characteristic of the TCB subassembly, a 3D drawing and prototype of three different TCBs with different designs are compared in Figs.
Following plate incubation, TCBS plates were checked for suspect colonies that were sucrose non-fermenting with a green or bluish-green color and a dark blue or green center approximately 3-5 mm long indicating the presence of V.

A minimum portion of the sample was taken with a platinum handle and seeded in duplicate by cross-streaking in boxes of Trypticase Soy Agar (TSA), Thiosulfate Citrate Bile Salts Sucrose Agar (TCBS) and selective media for the Pseudomonas group (Pseudomonas F Agar) The incubation time was 24-48 hrs at 34[degrees]C.
The swabs were plated onto five agars, from less selective to most selective, 5% sheep blood (Acumedia, CellBioSciences), marine salts enriched tryptose (Oxoid[TM]), McConkey agar with marine salts (Oxoid), thiosulfate-citrate-bile salts-sucrose agar (TCBS) (Acumedia, CellBioSciences), and marine salts agar with phenylethyl alcohol (Oxoid), and incubated at 30[degrees]C overnight.
Some tests that were carried out on each isolate to determine the presence of Vibrio cholerae after culturing in alkaline peptone water and thereafter subculturing in Thiosulphate Bile Salt Sucrose (TCBS) and a nutrient agar included Sulfide Indole Motility (SIM), Triple Sugar Iron (TSI), citrate utilization test, urease test, and oxidase tests.
One hundred microliters of the appropriate dilutions was surface-plated using a sterile cell spreader onto NA supplemented with 3% NaCl for enumeration of total bacteria and thiosulfate citrate bile salts sucrose agar (TCBS, Oxoid) to enumerate Vibrio species, respectively.
cholerae serotype Inaba (a gift from The Pasteur Institute of Iran) was grown on TCBS (thiosulfatecitrate-bile salts-sucrose agar, Merck, Germany) for 24 hours.
