For each cell line, growth was determined by both phase-contrast microscopy and demonstration of inclusions by thin-section electron microscopy. A variety of human (Hep-2, HEK, MRC-5, A549 and an Epstein Barr virus (EBV)--transformed human B-lymphoblastoid line), simian (Vero, LLC-MK2), and rodent (3T3, BHK) cell lines were used.
For thin-section electron microscopy, infected cells were fixed in cacodylate-buffered glutaraldehyde (2%), scraped from the flask, postfixed through increasing concentrations of ethanol (to 100% vol/vol), and then araldite embedded.
